Simple testing – the bacterial cells stain uniformly the color of the cells.
•
Negative staining –the bacteria is seen in clear spots against the dark background.

(a)Gram stain

Positive; purple stains by crystal violet.

Negative; no color change.
(b)Acid fast stain
•
Positive; Red to purple curved rods are observed, that occur in small groups which are
beaded.

(c)Capsule stain
•
Positive; clear halos zone against the dark background is observed.

Negative; no clear halos seen.
(d)Endospore stain
•
Positive; the spherical spores are located in the middle of the cell, bright green color is
observed.

Positive; piling of the stain on the flagella increase in thickness till are visible.
