SUPPLEMENTS
HYPOTHESIS
If a supplement label says it has x grams of protein, then the supplement contains the amount of
protein the label describes.

METHODS
1. In 4 small beakers, weigh out the appropriate amount of protein stock solution of known
concentration (20mg/mL). Label the beakers and put lids back on the correct containers.
2. Using the equation C1V1=C2V2, determine how to dilute the stock solution with water
to get the remaining known concentrations of standards. Mix the solutions thoroughly.
3. Insert a ‘blank’ test tube into the sample chamber and close the lid. The ‘blank’ is
concentration I for the three solutions.
4. Mix the standard solutions you prepare with Buiret’s Reagent as indicated for the color
indicator assay. Place the test tubes in the spectrophotometer and read absorbance at
565nm.
5. Record data in an excel chart and plot line graph.
6. Using the supplement in place of the protein stock solution (for this part, use three test
tubes) repeat steps 1 to 5.
