Abstract
This research paper details the neuro-cognitive disease Korsakoffs syndrome as it affects the
general population, the symptoms, etiology, and treatments currently employed to combat the
cognitive wasting caused by the syndrome. Comprehensively detailed starting with the history
and discovery of the illness in the late 19
th
century to the most recent genetic research into
mitochondrial etiologies and the links between these dysfunctions, the disease and other mental
health aberrations. Reviewed are case studies of those suffering from the genetic deficiencies
their symptomatology, treatment, and outcomes. Current treatment plans for both the alcohol
induced variant of the disease as well as the non-alcohol related illness, often the result of GI
tract issues, eating disorders, cancer and chemotherapy or undiagnosed malnutrition. The
principle causation of the disease is a thiamine deficiency which is often manifested in the vast
majority of the cases by alcoholism but as noted, there are other contributing factors in the
minority. A regimen of vitamin B
1
supplements, typically injections of 100-200mg/day are
sufficient in restoring much of the cognitive functions if caught early. Antioxidant therapies are
also prevalent treatments as they too, serve to restore brain function and help to repair previous
damage. For those that carry genetic abnormalities that manifest the syndrome, a treatment plan
may include a mixture of known mitochondrial supplements including Coenzyme Q10 100 mg,
L-arginine 2 gr vitamin C 1 gr, B1 100 mg, B2 10 mg, B3 30 mg, D3 2000 IU, E 200 mg). In
some cases the antipsychotic Cariprazine is also administered as 70% of those with the
SLC19A3 or 3243A mitochondrial dysfunction also suffer from some form of serious mental
health issue in addition to a thiamine deficiency. Public awareness and the stigma of mental
health stands in the way of effectively eradicating Korsakoffs syndrome for now.
