Differentiate cells by their maturation characteristics. Principle: Myelopoiesis is the process by which pluripotent stem mature and differentiate into mature leukocytes. The maturing leukocyte transition through different stages as it matures. Each maturation stage is characterize by unique morphological features which allows identification on a bone marrow smear. The stages identified from least mature to mature include myeloblast, promyelocyte, myelocyte, metamyelocyte, band and mature leucocyte. Maturation and differentiation through the different stages are regulated by different cytokines including GM-CSF, G-CSF, and M-CSF. There morphology and quantity of the different developing leukocyte precursors can be assessed in the bone marrow by studying key aspects of the developing cells such as size, cytoplasmic color, cytoplasmic granularity and nuclear changes. The information obtained provides a measure of red cell production in the bone marrow. Abstract
The objective of this experiment was to: identify the developmental morphology of each cell
in the series, identify each stage of maturation in each cell line, and differentiate cells in their
maturation characteristics. This report clearly explains the theory that was used in this
experiment. Besides, shows the materials and the supplies used in the experiment and the
procedure that was followed in doing the experiment. The results were presented in tables. The
maturation sequence was as follow; myeloblast stage, promyelocyte stage, myelocyte stage,
metamyelocyte stage, band stage, and mature leucocyte stage. In the monocytic maturation, the
sequence was as follow: eosinophil stage, basophil stage, monoblast stage, promonocyte stage,
mature monocyte stage, megakaryocyte stage, and plasma cell stage. Finally, the different
developmental stages were determined by the nuclear characteristics and the cytoplasmic
staining of each cell.
